Capabilities & Facilities
Streptonigrin is an antitumor antibiotic and a well-characterized ADC cytotoxin employed as an ADC payload in antibody-drug conjugates. Its cytotoxic mechanism involves intercalation into DNA and generation of reactive oxygen species (ROS), which induce single- and double-strand DNA breaks, leading to cell cycle arrest and apoptosis in proliferating tumor cells. The compound’s molecular structure allows efficient conjugation with antibody linkers for targeted delivery.
In antibody-drug conjugates, Streptonigrin is typically attached to monoclonal antibodies through cleavable or non-cleavable linker chemistries. The ADC remains stable in systemic circulation, preventing premature payload release, while intracellular enzymatic cleavage releases Streptonigrin within tumor cells. This approach ensures site-specific delivery of cytotoxic activity and supports precise antitumor mechanisms in targeted oncology applications.
Applications of Streptonigrin include its integration into ADCs directed against hematologic malignancies and solid tumors, including leukemia, lymphoma, breast cancer, and lung cancer. The payload is compatible with various linker chemistries, facilitating controlled intracellular release, stable antibody conjugation, and predictable pharmacokinetic behavior, all of which are essential for the design and production of ADCs with defined cytotoxic activity and therapeutic profiles.
What is Streptonigrin?
Streptonigrin is an antibiotic-derived cytotoxin used in ADC research. It intercalates DNA and generates reactive oxygen species, inducing DNA damage and apoptosis. Its mechanism of action makes it valuable as a payload for preclinical oncology studies targeting tumor cells.

Could you kindly advise how Streptonigrin acts as an ADC payload?
In ADCs, Streptonigrin is conjugated to tumor-targeting antibodies. After internalization, it intercalates DNA and produces oxidative stress, leading to cell cycle arrest and apoptosis, providing potent cytotoxic activity specific to target cells.
